Effect of In Vitro Irradiation on Platelet Aggregation and Ultrastructure


Abstract
This study was designed to evaluate the effect of in vitroirradiation on the platelet
function and ultrastructure. Platelet rich plasmas(PRPs) were prepared from 10 volunteer
blood donors and were irradiated with dosage varying from 1,000 to 6,000 rad by linear
accelerator. Platelet aggregation induced by ADP, epinephrine collagen and ristocetin
were evaluated in control and irradiated PRPs. Electron microscopical examination was
also performed after exposure to aggregating agents in control and irradiated platelets.
The results were obtained as follows.
1. Little differences were observed in aggregation patterns of control and irradiated
platelets following ADP, epinephrine, collagen and ristocetin treatment.
2. Ultrastructual features after irradiation in the platlets revealed loss of discoid shape,
pseudopod formation, numerous dilated vacuole formation and swelling in subperipheral
and sol-gel region. The changes in ultrastructure of organelle zone, which seemed to be
associated with platelet aggregation were negligidle.
3. These ultrastructural changes of the platelet were not correlated with radiation
dosage. Electron microscopical examination shows that the aggregation pattern of
various platelets was well observed in spite of postirradiated ultrastructural changes.
